Week 1: 7KB Plasmid Isolation
Introduction: In lab our first week back, my group and I constructed a plan for a 7KB Plasmid Isolation. A 7KB Plasmid is another plasmid inside of the Deinococcus cell. It is a stable and useful plasmid. Our attempt this semester is to extract and isolate the 7KB Plasmid from the Deinococcus cell. Procedure/Plan: In order to attempt one isolation and extraction, we would have to make 3mL of culture and media while having a minimum of 70mL of culture for one transformation. We likely will be using broth to perform this isolation. 1. Isolate 7KB from Deinococcus 2. Have proof of the actual 7KB that was isolated and use primers to cut at two specific places in the cell to make sure it is a 7KB 3. Once we are sure that the cell we extracted was the 7KB plasmid, we then isolate multiple and plenty of the plasmid.
